Can I use MS SOAP Toolkit 3.0 to call WSE 2.0 (example: myService.ashx) ?

By all means! WSE is a soap stack with WS-* implementation. You'd need
to write your own soap stack in Soaptoolkit to talk to WSE service.
Especially if you are using all the WS-* features in your WSE service.
