Block a Domain/URL

I'm trying to find a way to block all requests for a domain from going out.
Like when you use the Hosts file and redirect to 127.0.0.1(block) or the
actual IP(speedup?). Of course, I want to do this without using the Hosts
file and block URLs/Domains. Can anyone give me some general direction on
where to go to get more info on this. Thanks.

What you need is a firewall / packet filter...
